2)研究テーマ
地球上に生命が誕生して以来、約36億年が経ったといわれています。生命現象は熱力学第二法則(エントロピー増大の法則)に逆らう非常に不可思議な現象で、全宇宙で通用する物理・化学の法則を基盤とした「分子生物学」がこれだけ今日隆盛をきわめていても、尚、その本質は深い謎に包まれています。この連綿と続いてきた生命現象の根幹の一つに「生殖」があり、この根幹的な基本システムは、現在ではヒトも含めた殆どの生物種は「有性生殖」という形態をとっています。更に我々哺乳動物だけは受精卵を母体内寄生(=妊娠)させるという、極めて希有な生殖戦略をとり、その生命を今日まで連綿と繋げてきました。
地球上の多くの生命体が採った「有性生殖」という増殖戦略の根幹的な意義の解明を通じて、私たちの研究グループは「性差」の基本概念を構築することを目標として配偶子形成・受精・着床の分子機構の解明を中心に研究を進めています。また妊娠という生物として希有な戦略(=多くの生物種はリスクが高くその戦略を選択しなかったと推定される)を採ったが故に生じる様々な病態生理、その端的な身近な例として妊娠高血圧症候群などのヒト妊娠関連疾患の病態生理の本質をさまざまなアプローチを用いて探っています。
生殖生物学的な定義によれば、雄とは雄性配偶子(雌は雌性配偶子)を生み出す個体です。「雌鳥が卵を産む」のではなく、「卵を産むから雌鳥」と呼ばれています。このように性をその配偶子形成のための器官と考えると、そこには合目的的に特化した様々な分子機構が存在すると推定されます。近年、私たちは生殖細胞の分化の過程において特徴的な発現を示す uPAR/Ly6-like protein superfamily 分子群(GPI-アンカー型タンパク質)やバイセクト型GlcNAc糖鎖を中心とした研究を進めています。これらの分子はヒトを含めた哺乳動物に広く存在し、また極めてユニークな分子性状を持っていること、また発現分布様式から新しい生殖細胞の分化マーカーになりうる事などをこれまで明らかにしてきました。興味深いことにこれら分子の中には、ヒト組織の癌化に伴い発現してくることも近年明らかになっており、新しい腫瘍-精巣関連抗原として臨床的にも注目されてきております。これら分子の基礎的分析と生殖細胞形成・受精/着床における意義、また妊娠初期シグナルとしての卵(卵管を含めた)-免疫-内分泌ネットワークに焦点をあわせ、その生理機構や臨床医学上の応用に念頭にいれた研究を現在多角的に進めています。
